Mew can only be obtained through Nintendo Pokémon Events. The last event was the Toys R Us Mew event in 2006 but was only available in the USA. 10th Anniversary Mew was recently released in Japan, January 2010 to celebrate the anniversary of the Pokémon Gold/Silver games. Currently there is no news when the next Mew event will be available in North America.
You can get Mew by trading but just make sure to check its OT card info to verify its authenticity. Bulbapedia and Serebii have excellent page resources for all the event pokémon.
You can obtain Mewtwo several ways. The easiest way is it can be found in Cerulean Cave in HeartGold/SoulSilver in the Kanto region. Or you can import it to Pal Park from LeafGreen/FireRed after catching it in those games. Otherwise you will have to look for a potential trader who is willing to give up their Mewtwo.
